摘要:
利用纤维素的空间结构特点(海藻酸可与钙离子交联形成凝胶的性质),采用纤维素-海藻酸钠混合溶液滴入氯化钙溶液的包埋法固定糖化酶。考察纤维素-海藻酸钠用量比、纤维素-海藻酸钠混合溶液质量浓度、氯化钙溶液质量浓度等单因素对酶活力的影响。在单因素试验的基础上应用响应面优化试验,得到最优制备固定化糖化酶的工艺条件为:纤维素-海藻酸钠用量比例为1.5:1,纤维素-海藻酸钠混合溶液质量浓度为2.80%,氯化钙溶液质量浓度为2.77%,响应面优化结果验证良好。此外还进行了重复使用性实验,相比仅用包埋法制备的固定化糖化酶,包埋并且经过吸附酶液的固定化糖化酶重复使用性较好。
Abstract:
Cellulose,nad loose pore and long chain structure are favorable for the permeation and adsorption of macromolecules. When the calcium chloride solution was added to the mixed solution of cellulose and sodium alginate, cellulose and alginate can be cross-linked with calcium ions to form the gel. So glucoamylase could be immobilized by the carrier materials mixed with cellulose and sodium alginate. Some factors which affect the immobilized enzyme activity were considered, such as mix proportion for cellulose and sodium alginate, mass concentration of the mixed solution for cellulose and sodium alginate, and mass concentration of calcium chloride solution. The response surface methodology was carried out based on single factor test, and the optimal conditions for preparing immobilized glucoamylase were obtained as follows: the ratio of cellulose and sodium alginate was 1.5:1, the mass concentration of the mixed solution for cellulose and sodium alginate was 2.80%, the mass concentration of calcium chloride solution was 2.77%. The optimal conditions’ result of response surface methodology was tested successfully. In addition, the reusability test was conducted, and the results showed that the immobilized glucoamylase which adsorbed to the enzyme solution was better than unadsorbed and immobilized glucoamylase.
