针对当前电化学疗法中使用的传统化疗药物对人体正常组织毒副作用较大,中药成分对人体毒副作用小但作用效果弱等问题,选用中药单体成分二十碳五烯酸( EPA)作为抗癌药物,探究微秒脉冲电场对 EPA 细胞毒性效果的促进作用,以及两者联合作用对肺癌 A-549 细胞的 毒性 效果。 首先,研究不同场强 的脉 冲电场作用下细胞膜通透性变化情况,以及不同的药物浓度( 50 ~ 1 000 μmol / L) 、电场强度( 750 ~ 2 000 V / cm)对 A-549 细胞活性的影响,采用 PI 染色法检 测细胞 膜通 透性、CCK-8 法检测细胞活 性;其次,根据检测结果筛选用于后续实验的电场强度区间为 1 000 ~ 1 500 V / cm,EPA 浓度为 100 ~ 400 μmol / L;最后,设置电场联合药物组为实验组,空白对照组、纯药物组、纯电组为对照组,进 行了脉 冲电场联合 EPA 对 A-549 细胞毒性效果的实验研究,并且对脉冲电场联合 EPA 共同作用后培养 24 h(48 h)时A-549 细胞活性的变化情况进行了 对比分析。 研究表明:微秒脉冲电场能够显著增强 EPA 对 A-549 细胞的毒性效果,并且 EPA 浓度为 300 μmol / L 时,脉冲电场和 EPA 的联合作用效果最佳。 微秒脉冲电场对EPA 细 胞毒性的增强效 果可达 40%以上, 为后续中药成分干 预癌症的治疗提 供了重要的促进手段。
Abstract:
In view of the problems that traditional chemotherapeutic drugs used in current electrochemicaltherapy have large toxic and side effects on human normal tissues,and traditional Chinese medicine ingredi-ents have little toxic and side effects on human bodly , but have weak effects. Eicosapentaenoic acid (EPA ) , amonomer component of traditional Chinese medicine,was selected as an anticancer drug lo explore the promo-ting effect of microsecond pulse electric field on tlhe cyloloxic effect of EPA,and the toxic effect of the combi-nation of the two on lung cancer A-549 cells. Firstly ,the permeability of cell membrane under the aclion of pulsed electric field with different electric field strengths,and the effecls of different drug concentrations(50—1 000 u.mol/ L.) and electric field strengths (750—2 000 V/cm) on cell activity were studied. Pl stai-ning was used to detect cell membrane permeability , and CCK-8 assay was used to detect cell viability. Then ,according to the detection results , the electric field intensity range for subsequent experimenls was selected to)be 1 000—1 500 V/cm,and the EPA concenIration range was 100—400 umol/ L. Finally , the electric field/combined with the drug group was set as the experimental group,the blank conlrol group,the pure druggroup , and the pure electric group were set as the control group. The experimental study on the cytoloxicity ofpulsed electric field combined with EPA on A-549 cells was carried out,and the changes of A-549 cell viabili-lty al 24 h (48 h) after the combined action of pulsed electric field and EPA were compared and analyzed.Research shows thal microsecond pulsed electric field could significantly enhance the toxic effect of EPA on)A-549 cells, and when the concentration of EPA was 300 umol/l, the combined effect of pulsed electric field/and EPA was the best. The enhancement effect of microsecond pulsed electric field on EPA cytotoxicity couldreach more than 40% , which provided an important promotion method for the subsequent intervention of tradi-tional Chinese medicine ingredients in the treatment of cancer.
